SB743921 is a second generation, highly potent and active KSP inhibitor with a Ki of 0.1 nM for KSP (Eg5). It is 5-fold more potent against the ATPase activity of KSP than ispinesib. It has cytotoxic activity at less than 2 nM is a spectrum of tumor cell lines. It is negative in a mouse model of peripheral neuropathy in which paclitaxel is positive. [1] |
